Maclo®-100 contains aceclofenac BP 100 mg, a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain and inflammation in conditions such as osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis. It works mainly by inhibiting cyclo-oxygenase (COX) enzymes and reducing prostaglandin formation, which helps decrease inflammation and pain.

Pharmacology
Maclo-100 (Aceclofenac) is a non-steroidal agent with marked anti-inflammatory and analgesic properties. It is a potent inhibitor of the enzyme cyclooxygenase, which is involved in the production of prostaglandin. After oral administration, Maclo-100 (Aceclofenac) is rapidly and completely absorbed as an unchanged drug. Peak plasma concentration is reached approximately 1.25 to 3.00 hours after ingestion. Maclo-100 (Aceclofenac) penetrates into the synovial fluid where the concentration reaches approximately 57% of those in plasma. The mean plasma elimination half-life is around 4 hours. Maclo-100 (Aceclofenac) is highly protein-bound (<99%). Maclo-100 (Aceclofenac) circulates mainly as an unchanged drug, 4-Hydroxy Aceclofenac is the main metabolite detected in plasma. Approximately two-third of the administered dose is excreted via urine, mainly as hydroxymetabolite.
